After centrifugation, the cells were separated into two distinct fractions. A cell fraction obtained from the interphase of the centrifuge tube contained predominantly duct-like tubular structures, while the acini sedimented to the bottom. The ductal fraction was highly enriched in kallikrein, a ductal maker. To test the functional integrity of the prepared ductal cells, the effects of the membrane receptor agonists on intracellular free calcium concentration ([Ca^<2+>]i) were examined using the Ca^<2+> fluorescent indicator fura-2. Stimulation with carbachol, phenylephrine or isoproterenol increased significantly [Ca^<2+>]i in the ductal cells.
