The female hormones, estrogen and progesterone, play important roles in regulating the normal function of the human endometrium. They have also been found to affect the oral cavity. However, the effect of these female hormones on oral microbiota is yet unclear. Therefore, in this study, we aim to demonstrate the alteration in the oral microbiota caused by the administration of estrogen and progesterone using an ovariectomized mice model. Estrogen and progesterone were prepared in sesame oil and were administered subcutaneously to the experimental group mice daily for 20 days while the control group mice received the same volume of sesame oil only. DNA was extracted from the oral mucosa, and the oral microbiota was analyzed using next− generation sequencing. The differences in microbiota diversity, taxonomic abundance, and metagenomic functional prediction were analyzed. The progesterone administration caused a significant alteration in the α− and β−diversity of the oral microbiota. Taxonomy analysis revealed that the progesterone administration caused a significant increase in the proportion of Lactococcus spp. The metagenomic functional pathway prediction showed that the administration of estrogen and progesterone upregulated the activity of the propionic acid pathway, and the administration of progesterone upregulated the activity of the pyridoxal 5’−phosphate pathway. In this study, progesterone administration changed α− and β−diversity, and increased the proportion of Lactococcus spp. We also suggested that estrogen and progesterone administration increased pathways involved in propionate production, and progesterone administration increased pathways involved in pyridoxal 5’−phosphate. |
