内容記述 | The application of an immunohistochemical method in the detection of proliferating cells, that have incorporated thymidine analogue, 5-bromo-2-deoxyuridine (BrdU), was studied on cheek pouch mucosas of hamsters treated by three topical applications of 0.5% DMBA mineral oil. Incorporation of BrdU into newly synthesized DNA provides the basis for a simple technique to identify proliferating cells. BrdU was administered to hamsters by intraperitoneal injections at 6 hour intervals. Tissue was fixed in chilled acetone, embedded in soft paraffin, sectioned at 5μm. Deparaffinized tissue sections were incubated with an anti-BrdU antibody, and cells which had traversed the S-phase during the BrdU exposure period were identified immunohistochemically. Proliferation and migration of cheek pouch epithelial cells were identified by antibody staining after one, five, and nine exposures to BrdU. BrdU was mostly present in the nuclei of epithelial cells of the basal cell layer. The BrdU labeling Index (LI), or S-phase fraction, was determined by counting the number of BrdU-labeled cells in the tissue sections. The average LIs for non DMBA treated and DMBA-treated cheek pouch epithelia, administered a single dose of BrdU were 2.8% and 12.4% respectively. A higher proportion of labeled cells was presnt in the basal cell layer of the DMBA-treated animals than in that of the non-DMBA-treated animals. The ability to measure cell proliferation is important in the study of oral carcinogenesis. | |||||
