内容記述 | Permeabilized cells enabled us to gain access to the intracellular milieu required for exocytosis. Various permeabilization techniques have been applied to many secretory cells including pancreatic acini, however, studies with permeabilized salivary cells were very few. Therefore, I first detailed the preparation method of saponin-permeabilized parotid acini that were highly responsive to exogenous cAMP. Saponin-permeabilized parotid acini prepared in this manner were incubated with a wide variety of cAMP analogs, including site-selective cAMP analogs and the cAMP antagonist. The results suggest that the dissociation of cAMP-dependent protein kinase into its catalytic and regulatory subunits was essential for amylase exocytosis evoked by cAMP. Nevertheless, studies using protein kinase inhibitors and protein phosphatase inhibitors have shown that cAMP-dependent protein phosphorylation mediated by the catalytic subunit was not obligatory for exocytosis. The role of the regulatory subunit in amylase exocytosis remains to be elucidated. In this article, I also review recent studies concerning the effects of Ca^<2+>, C-kinase, and GTP-binding proteins on amylase exocytosis. | |||||
