The [Cl^-]_i was calculated from the SPQ fluorescence based on Englom\u0027s method. The resulting level of [Cl^-]_i of the rat parotid acinar cells was estimated to be 73.4±1.9mM (mean ± S.E., n=31). Stimulation with 10μM carba-chol, a muscarinic agonist, decreased the [Cl^-]_i to 50.1±3.1mM (n=12). The ED_\u003c50\u003e of carbachol necessary to change the [Cl^-]_i was 1.0μM. A similar decrease in [Cl^-]_i was elicited with 10nM of substance P or 10μM of phenylephrine, an α-adrenergic agonist. When the intracelular Ca^\u003c2+\u003e stores were previously depleted, no agonist-induced decrease in [Cl^-]_i was observed, suggesting that the decrease in [Cl^-]_i is induced by the increase in cytosolic Ca^\u003c2+\u003e concentration. No detectable change in [Cl^-]_i was observed when cells were stimulated with 1μM isoproterenol, a β-adrenergic agonist which induces the production of cyclic AMP. 内容記述 | Intracellular free chloride concentrations ([Cl^-]_i) in rat parotid acinar cells were determined with the Cl^- sensitive fluorescent indicator SPQ. The [Cl^-]_i was calculated from the SPQ fluorescence based on Englom's method. The resulting level of [Cl^-]_i of the rat parotid acinar cells was estimated to be 73.4±1.9mM (mean ± S.E., n=31).
